Cannabinoids and modulators of the endocannabinoid system affect specific mechanisms that are critical to the establishment and development of endometriosis. The aim of this study was to measure the systemic levels of endocannabinoids and related mediators in women with and without endometriosis and to investigate whether such levels correlated with endometriosis-associated pain. Plasma and endometrial biopsies were obtained from women with a laparoscopic diagnosis of endometriosis (n = 27) and no endometrial pathology (n = 29). Plasma levels of endocannabinoids (N-arachidonoylethanolamine [AEA] and 2-arachidonoylglycerol [2-AG]) and related mediators (N-oleoylethanolamine [OEA] and N-palmitoylethanolamine [PEA]), messenger RNA expression of some of their receptors (cannabinoid receptor type 1 [CB1], CB2, transient receptor potential vanilloid type [TRPV1]), and the enzymes involved in the synthesis (N-acyl-phosphatidylethanolamine-hydrolyzing phospholipase D [NAPE-PLD]) and degradation (fatty acid amide hydrolase 1 [FAAH]) of AEA, OEA, and PEA were evaluated in endometrial stromal cells. The systemic levels of AEA, 2-AG, and OEA were elevated in endometriosis in the secretory phase compared to controls. The expression of CB1 was higher in secretory phase endometrial stromal cells of controls versus endometriosis. Similar expression levels of CB2, TRPV1, NAPE-PLD, and FAAH were detected in controls and endometriosis. Patients with moderate-to-severe dysmenorrhea and dyspareunia showed higher AEA and PEA levels than those with low-to-moderate pain symptoms, respectively. The association of increased circulating AEA and 2-AG with decreased local CB1 expression in endometriosis suggests a negative feedback loop regulation, which may impair the capability of these mediators to control pain. These preliminary data suggest that the pharmacological manipulation of the action or levels of these mediators may offer an alternative option for the management of endometriosis-associated pain.

The days before menstruation are very complicated for women and there is a scientific explanation for your bad mood. Anandamide, a cannabinoid produced by our body, is highly dependent on estrogen. Specifically, an increase in cannabinoids and endocannabinoids decreases GnRH release, which ultimately also decreases progesterone. This holds up to general testing in which THC application reduces the circulating amount of both intermediary gonadal hormones, FSH and LH. Additionally, this holds across multiple species of mammals and even extends partially to amphibians!

According to WebMD, women can avoid discomfort, cravings that threaten their diet or the desire to kill someone thanks to consuming cannabis. After being medicated with cannabis, the woman will stabilize their estrogen levels and the terrible symptoms will disappear.

he Endocannabiniod System has a important role to play in female fertiltity. CB 1 Receptos have been shown to be present in the ovaries, uterine, endometrium, and the developing Embryo (McPartland 1999); These cannabis-like molecules also determine whether or not a fertilized egg implants in the uterus.
There is an increasing recognition that the endocannabinoid system is the crucial cytokine-hormone system regulating early human pregnancy. The synchronous development of the fertilized embryo and the endometrium to ensure timely implantation has been shown to be one of the pivotal steps to successful implantation. This development is thought to be regulated by a finely balanced relationship between various components of the endocannabinoid system in the endometrium, the embryo and the Fallopian tube. In addition, this system has also been shown to be involved in the regulation of the development and maturation of the gametes prior to fertilization.

Anandamide (AEA)
has been found to be present in the oviductal & follicular fluids ( fluids found in the fallopian tubes & the follicular antrum, the follicular antrum is the chamber in which ovarian follicles produce oocytes, or immature egg cells). CB 1 & 2 receptors have also been found in all stages of development of the oocyte itself. Mammalian sperm cells require a period of exposure to both male and female reproductive fluids in order to capacitate and able to fertilize a egg. The presence of Anandamide in the female reproductive fluids thus clearly assist in the capacitation of sperm cells on the journey to the fallopian tubes.  As well as assisting in capacitating the sperm cell and facilitating its journey to the fallopian tubes where fertilization takes place, Anandamide and the CB 1 receptors have a crucial roll to play in ovulation, as well as in the implementation of the fertilized gamete in to the endometrium, or the lining of the womb. Specifically it has been shown that high levels of Anandamide are necessary for successful ovulation, and that the lower levels are advantageous during the implementation of the gamete into the uterine lining ( Talatine et al, 2009 ) the same study also indicates that Anandamide is high at 4, & 5 weeks of gestation and drop to much lower levels during the 6th week. The concentration of anandamide in the oocytes has been found to strongly correlate with the maturation and quality of the oocytes (Taylor et al, 2010), which in turn determine the success of ovulation. During early gestation, successful embryo passage through the oviduct and implantation into the uterus require critical enzymatic control of the endocannabinoid levels (Taylor et al., 2010). We have previously described that AEA induces apoptosis of decidual cells, effects dependent on CB1 activation, involving p38 phosphorylation and a drop in the mitochondrial membrane potential (Fonseca et al., 2009Fonseca et al., , 2013) It is not clear exactly by what mechanism anandamide works to control oocyte maturation; the fact that high levels of anandamide are necessary at this time would appear to contradict the findings of the big Pharma sponsored trials, that the use of THC can disrupt oogenesis and ovulation.  The role of the endocannabinoid system in digestive health: cannabinoids are known to affect key physiological processes such as appetite and glucose metabolism, thereby potentially working to reduce the chances of obesity; obesity is strongly linked to reproductive health & Fertility, and the nutritional value of cannabis, especially raw,  (Kief Raw/Raw Kief/Raw Greens) is off the charts with Nutritional value, all Amino Acids, plus 4 newly discovered, cannabis is a natural medicine that can aid in the mimicking, enhancement, protectant, neuroprotectant, of all the bodily systems/pathophysiological processes, the entire hormonal system especially Anandamide as its a almost a identical twin but with more medicinal & nutritional punch & the list goes on and on.

Endocannabinoid system during development: emphasis on, developing unborn baby

The endocannabinoid system (ECS) including its receptors, endogenous ligands ("endocannabinoids"), synthesizing and degradating enzymes, and transporter molecules has been detected from the earliest embryonal stages and throughout pre- and postnatal development; endocannabinoids, notably 2-arachidonoylglycerol, are also present in maternal milk. During three developmental stages, (1) early embryonal, (2) prenatal brain development, and (3) postnatal suckling, the ECS plays an essential role for development and survival. During early gestation, successful embryonal passage through the oviduct and implantation into the uterus require critical enzymatic control of the endocannabinoids. During fetal life, endocannabinoids and the cannabinoid CB(1) receptor are important for brain development, regulating neural progenitor differentiation and guiding axonal migration and synaptogenesis. Postnatally, CB(1) receptor activation by 2-arachidonoylglycerol appears to play a critical role in the initiation of milk suckling in mouse pups, possibly by enabling innervation and/or activation of the tongue muscles. Perinatal manipulation of the ECS, by administering cannabinoids or by maternal marijuana consumption was a common practice in the past.

 With all the methods that cannabis was utilized for it's own healing properties, never mind the ECS regulatory functions, some of the more recorded cases from the past is for the aid in nausea, Pain, migraines & Inflammation, ( history  or history timeline ) all of which are a common ordeal throughout a pregnancy. A significant cause of brain injury in the neonatal period is perinatal hypoxic-ischemic event—when the flow of oxygen and blood to the infant brain is interrupted due to asphyxia, often during childbirth. This devastating condition leads to the death of 15-20% of children diagnosed, and to severe neurological impairment—including epilepsy, cerebral palsy, motor dysfunction and hyperactivity—in a further 25%. The developing brain is far more susceptible to hypoxic-ischemic events than the adult brain, as it contains a high concentration of blood vessels and higher water content, and the potential for damage-inducing events such as haemorrhage is greater.
Immediately following hypoxic-ischemic brain injury, a set of specific cellular mechanisms including increased production of glutamate is set in motion, triggering cell damage and finally leading to excitotoxicity (a type of cell death caused by excessive glutamatergic signalling).

Estrogen Levels & the Endocannabinoid System
